No, you only need to conjure once, then let it burn. You don't have to relight unless you want to because the candles are reusable for other situations.

When you light any conjure candle on its day you can leave it lit until it burns out or you can snuff it out after 6 hours. The highest energy hours to lite them are 6am 1pm and 8pm, but as long as their lit on there day you will be fine.
